Summary
This gene encodes a dual functional inositol kinase. The encoded enzyme converts inositol hexakisphosphate to diphosphoinositol pentakisphosphate and diphosphoinositol pentakisphosphate to bis-diphosphoinositol tetrakisphosphate. This protein may be important for intracellular signaling pathways. Alternate splicing results in multiple transcript variants. A pseudogene of this gene is found on chromosome 15.[provided by RefSeq, Jun 2010

LÄHDE:  p53-mediated apoptosis requires inositol hexakisphosphate kinase-2

Abstract
Inositol pyrophosphates have been implicated in numerous biological processes. Inositol hexakisphosphate kinase-2 (IP6K2), which generates the inositol pyrophosphate, diphosphoinositol pentakisphosphate (IP7), influences apoptotic cell death. The tumor suppressor p53 responds to genotoxic stress by engaging a transcriptional program leading to cell-cycle arrest or apoptosis.

We demonstrate that IP6K2 is required for p53-mediated apoptosis and modulates the outcome of the p53 response. Gene disruption of IP6K2 in colorectal cancer cells selectively impairs p53-mediated apoptosis, instead favoring cell-cycle arrest. IP6K2 acts by binding directly to p53 and decreasing expression of proarrest gene targets such as the cyclin-dependent kinase inhibitor p21.

Among the inositol phosphates, inositol 1,4,5-trisphosphate is best known for its release of intracellular calcium (1). The inositol pyrophosphates (IPPs), synthesized by inositol hexakisphosphate kinases (IP6Ks), regulate numerous processes including chemotaxis (2), telomere length (3, 4), endocytic trafficking (5), exocytosis (6), and apoptosis (7, 8).

The principal inositol pyrophosphate, diphosphoinositol pentakisphosphate (5-PP-IP5), here designated IP7, is generated by three IP6 kinases that are the products of three separate genes (9).

Another isomer of IP7, 3-PP-IP5, is synthesized by a distinct enzyme, Vip1, and regulates cell shape, growth, and phosphate disposition of yeast (10, 11).
IP6K1 has been directly implicated in vesicular trafficking and tissue growth, because IP6K1-deleted mice manifest diminished insulin release, slowed growth, and defects in spermiogenesis (12). IP6K2 selectively impacts cell death, because its overexpression sensitizes cells to diverse apoptotic stimuli such as DNA damage, hypoxia, hydrogen peroxide, and interferon-β; knockdown of IP6K2 but not IP6K1 or IP6K3 diminishes sensitivity to such stimuli (7, 8, 13, 14).

Recently, Lindner and coworkers (15) developed IP6K2 knockout mice that are predisposed to invasive aerodigestive tract carcinoma driven by chemical carcinogenesis, and fibroblasts from the mice resist γ-irradiation. The mechanism by which IP6K2 regulates cell death has not been established.

We now show that IP6K2 is required for p53-mediated apoptosis and acts by binding p53 and selectively diminishing expression of its pro-cell-cycle arrest targets.

Organelle Crosstalk in Membrane Dynamics and Cell Signalling
New molecular mechanisms of inter-organelle lipid transport

  • Abstract
  • Lipids are precisely distributed in cell membranes, along with associated proteins defining organelle identity. Because the major cellular lipid factory is the endoplasmic reticulum (ER), a key issue is to understand how various lipids are subsequently delivered to other compartments by vesicular and non-vesicular transport pathways.
  •  Efforts are currently made to decipher how lipid transfer proteins (LTPs) work either across long distances or confined to membrane contact sites (MCSs) where two organelles are at close proximity.
  • Recent findings reveal that proteins of the oxysterol-binding protein related-proteins (ORP)/oxysterol-binding homology (Osh) family are not all just sterol transporters/sensors: some can bind either phosphatidylinositol 4-phosphate (PtdIns(4)P) and sterol or PtdIns(4)P and phosphatidylserine (PS), exchange these lipids between membranes, and thereby use phosphoinositide metabolism to create cellular lipid gradients.
Lipidien vaihto on todennäköisesti  laajalle levinnyt mekanismi , jota hyödyntää muutkin lipidien kuljettajaproteiinit  jakaen tehokkaasti lipidejä eri organellikalvojen  kesken.
  •  Lipid exchange is likely a widespread mechanism also utilized by other LTPs to efficiently trade lipids between organelle membranes.
  •  Finally, the discovery of more proteins bearing a lipid-binding module (SMP or START-like domain) raises new questions on how lipids are conveyed in cells and how the activities of different LTPs are coordinated.

PI -PthOH kierto PM-ER vyöhyke, Nir2-kuljettajaproteiini , ALS

Phosphatidylinositol  (PI)  and phosphatidic acid (PA) transport between the ER and plasma membrane during PLC activation requires the Nir2 protein.

Tiivistelmä  Suomennettu 24.6. 2017

Abstract

Rajoittuneen  plasmamebraanilipidi PIP2- altaan PLC- välitteinen hydrolyysi vaatii täydentämistä laajasta PI altaasta ja siinä toimii kinaasit PI4-K- ja PI4P-5K.
Koska PI molekyylit syntetisoituvat endoplasmisessa retikulumissa (ER)  ja PIP2 generoituu plasmakalvossa, on oletettu, että  PI molekyyliä siirtävät proteiinit (PITP) antavat keinon tämän lipidin siirtämisfunktioon.   Tuoreet tutkimukset ovat tunnsitaneet PITP-proteiinin Nir2 olevan tärkeä PI- molekyylin kuljettamiseksi endoplasmiseseta retikulumista  plasmakalvoon.havaittiin myös , että Nir2 proteiini vaaditaan kuljetatmaan  fosfatidihappoa PtdOH  plasmakalvosta endoplasmiseen retikulumiin.   Nämä löydöt  ovat valottaneet PI ja PAmolekyylien  ei-vesikulaarista  kuljetusta  ER-P; kontaktivyöhykkeessä, josta on mahdollinen linkkiytyminen hyvin tuhoisiin ihmistauteihin.

Jos Nir2-kuljettajaproteiini puuttuu soluista ,PLC-aktivaatio johtaa  fosfatidaatin (PtdOH) kertymiseen plasmakalvoon ja PI-synteesi vaikeutuu vakavasti.
Lepäävissä soluissa Nir2 lokalisoituu endoplasmiseen verkostoon tekemällä interaktion FFAT domaaniin  ER-sitoutuneissa VAMP-assosioituneissa proteiineissa  VAP-A ja VAP-B.
VAP-B mutaatiot joita on tunnsitettu amyotroofisessa lateraaliskleroosissa (ALS)  eli Lou-Gehrigin taudissa , aiheuttavat VAP-B-proteiinin aggrekaatioita, jotka sitten haittaavat sitoutumalla useisiin proteiineihin joihin kuuluu Nir2.
  • Phospholipase C (PLC)-mediated hydrolysis of the limited pool of plasma membrane (PM) phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] requires replenishment from a larger pool of phosphatidylinositol (PtdIns) via sequential phosphorylation by PtdIns 4-kinases and phosphatidylinositol 4-phosphate (PtdIns4P) 5-kinases.
  •  Since PtdIns is synthesized in the endoplasmic reticulum (ER) and PtdIns(4,5)P2 is generated in the PM, it has been postulated that PtdIns transfer proteins (PITPs) provide the means for this lipid transfer function.   Recent studies identified the large PITP protein, Nir2 as important for PtdIns transfer from the ER to the PM.   It was also found that Nir2 was required for the transfer of phosphatidic acid (PtdOH) from the PM to the ER.
  •  In Nir2-depleted cells, activation of PLC leads to PtdOH accumulation in the PM and PtdIns synthesis becomes severely impaired.  In quiescent cells, Nir2 is localized to the ER via interaction of its FFAT domain with ER-bound VAMP-associated proteins VAP-A and-B.
  •  After PLC activation, Nir2 also binds to the PM via interaction of its C-terminal domains with diacylglycerol (DAG) and PtdOH. Through these interactions, Nir2 functions in ER-PM contact zones.
  • Mutations in VAP-B that have been identified in familial forms of amyotrophic lateral sclerosis (ALS or Lou-Gehrig's disease) cause aggregation of the VAP-B protein, which then impairs its binding to several proteins, including Nir2.
  •   These findings have shed new lights on the importance of non-vesicular lipid transfer of PtdIns and PtdOH in ER-PM contact zones with a possible link to a devastating human disease.

VIII. Protein Phosphatase-Directed Therapeutics for the Prevention and Treatment of Alzheimer’s Disease

According to the National Institute on Aging, there are more than 5 million Americans who suffer from AD. Yet, despite a desperate need, development of an effective treatment for AD has been a major challenge. Four drugs are currently approved by the FDA to treat cognitive deficits in AD. Three of them are acetylcholinesterase inhibitors, which combat the loss of acetylcholine caused by the death of cholinergic neurons, while the other is a noncompetitive NMDA receptor antagonist, which inhibits overactivation of NMDA receptors by glutamate.1 None of these drugs halts progression of the disease. Significant effort has been put into development of inhibitors of Aβ production.233 However, targeting γ-secretase with nonselective γ-secretase inhibitors has deleterious effects on health, likely because γ-secretase also cleaves other substrates, such as Notch, which are essential for normal biological function.234 This limitation is highlighted by the recent failure of the γ-secretase inhibitor semagacestat in Phase III clinical trials.235 Given the lack of available drugs to treat AD, it is imperative to identify novel molecular processes that might be amenable to targeting through new drugs. Protein phosphatases, including PP2A, and STEP may be potential therapeutic targets for AD. It may also be possible to use a strategy that would combine one or more regulator of a protein phosphatase with kinase inhibitors that would provide a complementary approach to control of, for example, hyperphosphorylation of tau.

PS: Kappale on pitkä ja siinä on paljon asiaa. hyperfosforylaatiossa on yksi  seikka mitä voisi myös ottaa pinsettiin: orgaanisen ja epäorgaanisen  fosfaatin suhde kehossa, sillä siinäkin on moduloimisen varaa  dietääriä tietä.-   Tämä suhde on  nykyaikana käänteinen verrattuna  entisaikojen ravinnon  aiheuttamaan  fosfaattisuhtgeeseen.
