Inositolin määrä maidossa

Abstract

The gaschromatographic method was used for the quantitative analyses of inositol in milk. The content of inositol in human and cow's milk at the different lactation periods was determined. The content of total myoinositol in human milk was 32.7 plus or minus 15.2 mg/100 ml in colostrum, 17.8 plus or minus 1.9 mg/100 ml in transitional milk, and 14.9 plus or minus 3.1 mg/100 ml in mature milk.

In cow's milk, it was 10.6 plus or minus 1.0, 7.0 plus or minus 1.1, and 4.1 plus or minus 1.0 mg/100 ml, respectively. These values were very similar to those obtained by the microbiological method. The presence of lipid-bound myoinositol in both kinds of milk was confirmed and the content was 0.22 plus or minus 0.09 mg/100 ml in human milk and 0.36 plus or minus 0.10 mg/100 ml in cow's milk. A small amount of scylloinositol was found in both human and cow's milk, while dextroinositol was not found in either.

Abstract

Breast milk delivers nutrition and protection to the developing infant. There has been considerable research on the high-molecular-weight milk components; however, low-molecular-weight metabolites have received less attention. To determine the effect of maternal phenotype and diet on the human milk metabolome, milk collected at day 90 postpartum from 52 healthy women was analyzed by using proton nuclear magnetic resonance spectroscopy. Sixty-five milk metabolites were quantified (mono-, di-, and oligosaccharides; amino acids and derivatives; energy metabolites; fatty acids and associated metabolites; vitamins, nucleotides, and derivatives; and others). The biological variation, represented as the percentage CV of each metabolite, varied widely (4-120%), with several metabolites having low variation (<20 b="" glutamate="" including="" lactose="" urea="">myo-inositol,
and creatinine. Principal components analysis identified 2 clear groups of participants who were differentiable on the basis of milk oligosaccharide concentration and who were classified as secretors or nonsecretors of fucosyltransferase 2 (FUT2) gene products according to the concentration of 2'-fucosyllactose, lactodifucotetraose, and lacto-N-fucopentaose I.
 Exploration of the interrelations between the milk sugars by using Spearman rank correlations revealed significant positive and negative associations, including positive correlations between fucose and products of the FUT2 gene and negative correlations between fucose and products of the fucosyltransferase 3 (FUT3) gene. The total concentration of milk oligosaccharides was conserved among participants (%CV = 18%), suggesting tight regulation of total oligosaccharide production; however, concentrations of specific oligosaccharides varied widely between participants (%CV = 30.4-84.3%). Syö kuituja, kehottavat ravitsemussuositukset. Nyt perään voi lisätä: tai sinut syödään sisältäpäin.
Mikäli kiltit ja hyödylliset suolistomikrobimme eivät saa tarpeekseen ravintokuituja, ne alkavat natustaa paksusuolen pintaa suojeleva limaa. Suolen seinämä voi rapautua tästä syystä niin, että se tulehtuu helposti.
Mikrobiologi Eric Martens Michiganin yliopistosta kollegoineen selvitti kuitujen merkitystä hiirillä. Niihin istutettiin 14 bakteerilajia, jotka kasvavat ihmisen suolistossa.
Sitten ryhmä tutki, mitä jyrsijöiden suolistossa tapahtuu, kun niiden ravinnon kuitupitoisuus muuttuu. He altistivat hiiret myös bakteereille, jotka tekevät niiden suolistossa samaa kuin Escherichia coli -bakteeri ihmisissä eli aiheuttavat tulehdusta ja ripulia.
Kun hiiret söivät ruokaa, joka sisälsi noin 15 prosenttia kuituja, niiden suoliston limakerros pysyi ennallaan ja tulehdusta aiheuttavat taudinaiheuttajat aisoissa.
Sen sijaan kuiduton ruoka usutti suolistomikrobit liman kimppuun muutamassa päivässä.
Tutkijat lisäsivät kokeeksi ruokaan prebiootteja, joita käytetään monissa elintarvikkeissa. Ne johtivat hiirissa samanlaiseen liman katoamiseen kuin kuiduton ruoka.
Mikrobikirjo muuttui jopa päivittäin sen mukaan, mitä hiiret söivät. Jotkin bakteerilajit pärjäsivät paremmin vähäkuituisissa, toiset runsaskuituisissa oloissa.
Neljä kuiduitta kukoistavaa mikrobilajia olivat ainoita, jotka tuottavat suolen liman proteiineja hajottavia entsyymejä.
Mitä vähemmän kuituja hiiri söi, sitä ohuempi limaa tuottavien solujen kerros oli. Hento limapeite ei suojannut taudinaiheuttajilta.
”Vaikka tämä tutkimus tehtiin hiirillä, tulos vahvistaa sen, mitä ravitsemustieteilijät ovat toitottaneet vuosikymmeniä. Kannattaa syödä paljon luonnollisia kuituja. Ruokavalio vaikuttaa suoraan suolistomikrobeihin ja sairastumisalttiuteen”, Martens sanoo tiedotteessa.
Ravintokuituja on runsaasti täysjyväviljassa, palkokasveissa, pähkinöissä ja siemenissä.

KOMMENTTI NI: Tähän voi vielä lisätä : keliaakikon pitää syödä ne kuidut gluteenittomina, siis muita kuituja kuin kotimaisesta viljasta. keliaakikoille on onneksi tehty gluteenittomia kuituvalmisteita kuten FIBREX sokerijuurikaskuitu, PO-fiber, perunakuitu ja HUSK psylliumsiemenkuitu.
Näistä FIBREX on  mukava käyttää esim vohveli ja lättytaikinassa tai puuroissa lisänä.
PO-fiber ja HUSK on sopivia leivontaan.

After antigen receptor engagement, the membrane lipid phosphatidylinositol-4,5-bisphosphate (PtdIns(4,5)P2) — which has a membrane-embedded lipid moiety and a cytoplasm-exposed charged inositol phosphate (InsP) moiety — can either be converted into PtdIns(3,4,5)P3 (referred to here as PtdInsP3) by phosphoinositide 3-kinases (PI3Ks) or into the second messengers diacylglycerol (DAG) and soluble Ins(1,4,5)P3 (referred to here as InsP3) by phospholipase Cγ1 (PLCγ1) or PLCγ2.

Phosphatase and tensin homologue (PTEN) counteracts PI3Ks.

 SH2-domain-containing inositol-5-phosphatase 1 (SHIP1) and SHIP2 convert PtdInsP3 into PtdIns(3,4)P2. ( reaktio vain yhteen suuntaan).

 Primarily through their InsP headgroups, PtdIns(4,5)P2, PtdInsP3 and PtdIns(3,4)P2 can bind to pleckstrin-homology (PH) domains in certain proteins and recruit them to membranes.

(Yleisesti tunnettua!)    InsP3 mobilizes Ca2+ but can also be phosphorylated at position 3 to form Ins(1,3,4,5)P4 (referred to here as InsP4) by InsP3 3-kinases, including ITPKA, ITPKB, ITPKC and InsP multikinase (IPMK)11, 17, 18, 23, 34, 35.


Owing to its similarity to the PtdInsP3 headgroup, InsP4 can bind to the PH domains of certain PtdInsP3-binding proteins and
 promote (in the case of IL-2-inducible T cell kinase (ITK) and tyrosine kinase expressed in hepatocellular carcinoma (TEC))
or inhibit (in the case of AKT and possibly Ras GTPase-activating protein 3 (RASA3)) the binding of these proteins to PtdInsP3.

In CD4+CD8+ thymocytes, InsP4-mediated promotion of PtdInsP3 binding to the ITK and/or TEC PH domains establishes a feedback loop of PLCγ1 activation.

 In neutrophils, InsP4-mediated inhibition of the binding of PtdInsP3 or PtdIns(3,4)P2 to the PH domain of AKT may limit the membrane recruitment of AKT.

InsP4 can also inhibit RASA3 binding to PtdIns(4,5)P2 or PtdInsP3 (Refs 18, 21). Whether this occurs in leukocytes is unknown. Red boxes denote enzymes with demonstrated physiological relevance in leukocytes. R, fatty-acid side chain.
